RHU LUMIERE

A University-Affiliated Hospital Research Project to Study the Role of the Gut Microbiome in the Response to Cancer Immunotherapies in Bronchial Cancers.

Lung cancer causes nearly 30,000 deaths per year in France. Therapies based on blocking T-cell inhibitory receptors (ICBs), such as anti-CTLA-4 and anti-PD(L)1, can prolong survival in some patients. However, primary resistance and severe autoimmune side effects have been observed. In recent years, several research teams, including ours, have shown that the gut microbiota plays an important role in the anti-tumor immune response.

The goal of LUMIERE is to highlight the role of gut microbiota imbalance in the response to ICBs in humans. It also aims to determine whether modifying the gut microbiota could restore the antitumor response.

Objectives of the LUMIERE Project

  1. To develop new diagnostic tools, based on metagenomic signatures of the gut microbiota, that can predict or forecast response to or resistance against ICBs in order to avoid inappropriate prescribing for non-responders.
  2. To develop new therapeutic solutions such as “oncobiotics” (immunogenic commensals) and “bioactives” (derivatives, metabolites) to stimulate the activity of ICBs and even overcome primary resistance. The estimated economic benefits extend far beyond applications for lung cancer alone, encompassing all types of cancer treated with ICBs.

Funding

9 million euros

The LUMIERE project is receiving €9 million in funding from the second wave of the Investment for the Future Program (PIA2), which is administered by the National Research Agency (ANR). This program aims to support innovative, large-scale research projects in the field of health.

The Participants in the LUMIERE Project

Bringing together academic experts in oncology, immunology, microbiology, metagenomics, and culturomics, an innovative biotechnology company, and two global leaders in the pharmaceutical industry, the consortium’s work represents a departure from current clinical practices. The project, led by Laurence Zitvogel (Gustave Roussy) and the DHU TORINO (Gustave Roussy Institute, AP-HP, Paris-Sud University, Inserm), will run for 60 months.
