Summary
Our platform makes its expertise available to basic and clinical research teams at Gustave Roussy and Paris-Saclay, as well as to national and international partners. It offers high-resolution imaging approaches tailored to oncology research, organized around two complementary areas: cellular imaging, dedicated to the analysis of subcellular structures and their dynamics, and molecular imaging, focused on the study of DNA, RNA, chromatin, and nucleoprotein complexes.
Applications include the intracellular tracking of therapeutic agents via various internalization pathways, the study of cellular function (intracellular trafficking, organelle organization, metabolic dysfunctions, cell death), as well as the analysis of nuclear structures (organization and relocalization of chromatin, interactions with the lamina, formation of condensates and membrane-less organelles, nucleolus morphology). The platform possesses specific expertise in nucleic acid imaging, enabling the extraction of nucleic acids from cells and the analysis of DNA replication and repair intermediates, the characterization of R-loops, as well as the study of large chromosomal regions, particularly centromeres and telomeres. It develops biochemical and structural approaches to analyze nucleoprotein complexes targeted by inhibitors, RNA folding, and liquid-liquid phase separation (LLPS).
The platform is equipped with advanced instruments (Tecnai 12 and Zeiss 902/912 electron microscopes, an ultramicrotome, cryosubstitution, AFM) as well as preparation and analysis tools (validated antibodies, RNA probes, CryoEM with Vitrobot, and access to Titan Kryos).
