Summary

Objectives and Challenges

Team 1, “Antitumor Immunity of T Cells and Cancer Immunotherapy,” studies the mechanisms that control the efficacy of antitumor immune responses, particularly those mediated by T cells, NK cells, and CAR-T cells. Its goal is to understand the causes of resistance to immunotherapies in order to identify new predictive biomarkers and develop more effective therapeutic strategies for adult and pediatric patients.

Research Areas / Major Projects

The research is organized around four areas: study of resident memory T cells and resident NK cells; impact of the tumor microenvironment on effector cells; personalized T-cell therapies for virus-associated cancers; and optimization of CAR-T cells in solid tumors. The team combines flow cytometry, single-cell transcriptomics, dynamic imaging, in vivo models, and ex vivo 3D models of human tumors, along with spatial analyses of the tumor microenvironment.

Major Findings, Prospects, and Impacts

The team has contributed to a better understanding of the determinants of the efficacy of antitumor responses and immunotherapies. Their work has focused in particular on the role of resident T cells, the identification of Nrp-1 as a new immunological target, the development of vaccines based on TEIPP antigens, and the use of memory T stem cells in cell therapy. It has also studied the impact of several components of the tumor microenvironment—the extracellular matrix, macrophages, hypoxia, p53 status, and mitochondrial metabolism—on the activity of T cells and CAR-T cells. These findings open up new avenues for improving immunotherapies, guiding biomarker-based treatments, and designing new cellular or combination therapeutic approaches.
