Summary
Objectives and Challenges
The team is studying tumor-associated macrophages (TAMs), key players in the tumor microenvironment (TME), as well as other subpopulations of myeloid cells. Despite the success of immunotherapies targeting immune checkpoint inhibitors (ICIs), TAMs—which are heterogeneous and poorly understood—limit their effectiveness. Their diversity (embryonic or monocytic origin, duration of residence in the tumor) and their pro- or anti-tumor roles remain to be elucidated in order to develop targeted and personalized therapies.
Research Areas / Main Projects

Mapping of myeloid cells: Use of unique mouse models to trace the heterogeneity of myeloid cells (ontogeny, residence time).

Integration of human data: Multidimensional analysis (mRNA, proteins, epigenetics, spatial localization) of dynamic cohorts to identify new therapeutic targets.

3D modeling: Study of macrophages in pediatric cancers using organoids/tumoroids derived from iPS cells, for personalized approaches (“patient avatar”).
Key findings, future directions, and impacts

Results: Discovery of the developmental origin of macrophages (3 distinct waves) and their adaptation to tissue niches, applied to the TME.

Future Directions: Identifying myeloid targets for new immunotherapies, taking into account the diversity of TAMs and their location within the tumor.
