Summary
Themes, Objectives, and Challenges
Our team is studying megakaryopoiesis—the process of platelet production—both in normal and pathological contexts related to myeloproliferative neoplasms (MPNs) and hereditary thrombocytopenias (HTs). Our main objectives are: 1) to understand the initiation of MPNs by determining the age of onset of mutations and the dynamics of mutated CSH; 2) to study normal and pathological megakaryopoiesis by focusing on the pathophysiology of HT, MPDs (at the stages of myeloproliferation and myelofibrosis), and acute megakaryoblastic leukemia; 3) to study the mechanisms of transformation into acute myeloid leukemias (AMLs) secondary to NMPs, hereditary factors (neutropenia, thrombocytopenia, Trisomy 21, 14q32 duplication), or to chemotherapies used in solid tumors; 4) develop innovative selective therapies for MPNs and AMLs, as well as blood transfusion products (platelets produced from iPSCs).
Results, Prospects, and Scientific, Clinical, and Societal Impacts
Our work has helped elucidate the timing of the emergence of JAK2V617F and mutated CALR mutations, their effects on hematopoietic stem cells, and the role of genetic factors in the development of NMPs. It has also helped elucidate the mechanisms by which pathological megakaryocytes induce myelofibrosis and TH. Furthermore, our research has led to a better understanding of leukemic transformation associated with TP53 mutations, trisomy 21, and certain TH. Finally, promising advances have been made in the development of innovative therapeutic approaches, including selective therapies for NMPs and platelet production technologies for clinical use. Our research has significant scientific impact, opens up new therapeutic avenues, and improves the clinical management of hematologic diseases.
